Best runes for the selected Kindred build:
Lamb and Wolf mark champions and monster camps, respectively. Taking down the marked targets permanently empower Kindred's abilities, and give Lamb bonus range.
Lamb dashes in the target directio, gaining bonus attack speed and fires up to three arrows at nearby targets dealing physicla damage.
Kindred passively build stacks as they move and attack that heal once full. Activate to send Wolf to a target location where he will attack enemies with the area.
Cripple a target, slowing them by 50% for 1 second. If Lamb attacks a target twice within 4 seconds, a third attack within the next 4 seconds will send Wolf at the target to deal additional damage.
Lamb creates a circle around her in which all units cannout be reduced below 10% of their max health. Units who reach that threshold cannot be healed in the area. All units are healed once it ends.

Despite marks being super important, do not tunnel vision on them where you put yourself in bad situations to get them.. The enemy team can see your marks so it's not hard for them to collapse on you since they know where you will be going.
